چکیده

The common use of Agrobacterium as a gene vector for plants has somewhat ob-scured the fact that this bacterium remains an important plant pathogen. Pathogenic strainsof the genus Agrobacterium cause unorganized tissue growth called crown gall or profuseabnormal root development called hairy root. Agrobacterium tumefaciens induces galls onroots and crowns of several fruit and forest trees and ornamental plants. A. vitis is responsi-ble for the crown gall disease of grapevine, while A. rhizogenes induces abnormal rootingon its hosts. Plants tissues that become diseased undergo physiological changes resulting inweak growth, low yields or even death of the entire plant. Tumors originate from dividingplant cells, e. g. from cambium. Thus the cambial region becomes unable to differentiateinto efficient phloem and xylem elements leading to deficient nutrient transport. Symptomsmay appear on roots, crowns and aerial parts of attacked plants (Figure 1-1). Tumors areusually comprised of unorganized tissue, but sometimes they differentiate into roots orshoots. This depends on the host plant, the position on the infected plant or the inducingbacterium (Figure 1-2). As indicated by several reviews, crown gall has been a worldwideproblem in agriculture for over hundred years (Moore and Cooksey, 1981; Burr et al., 1998;Burr and Otten, 1999; Escobar and Dandekar, 2003). 2 Léon Otten, Thomas Burr and Ernö Szegedi

Agrobacterium-mediated plant transformation: the biology behind the "gene-jockeying" tool.

Agrobacterium tumefaciens and related Agrobacterium species have been known as plant pathogens since the beginning of the 20th century. However, only in the past two decades has the ability of Agrobacterium to transfer DNA to plant cells been harnessed for the purposes of plant genetic engineering. Since the initial reports in the early 1980s using Agrobacterium to generate transgenic plants, s...

Agrobacterium-mediated genetic transformation of plants: biology and biotechnology.

Agrobacterium-mediated genetic transformation is the dominant technology used for the production of genetically modified transgenic plants. Extensive research aimed at understanding and improving the molecular machinery of Agrobacterium responsible for the generation and transport of the bacterial DNA into the host cell has resulted in the establishment of many recombinant Agrobacterium strains...

From the tumor-inducing principle to plant biotechnology and its importance for society.

This dialogue was held between the Guest Editors of the Special Issue on "Plant Transgenesis" of the Int. J. Dev. Biol. and Marc Van Montagu. Research in the group of Marc Van Montagu and Jeff Schell in the 1970s was essential to reveal how the phytopathogenic bacterium Agrobacterium tumefaciens transfers DNA to host plants to cause crown gall disease. Knowledge of the molecular mechanism under...

The roles of bacterial and host plant factors in Agrobacterium-mediated genetic transformation.

The genetic transformation of plants mediated by Agrobacterium tumefaciens represents an essential tool for both fundamental and applied research in plant biology. For a successful infection, culminating in the integration of its transferred DNA (T-DNA) into the host genome, Agrobacterium relies on multiple interactions with host-plant factors. Extensive studies have unraveled many of such inte...

Agrobacterium tumefaciens as an agent of disease.

Twenty-six years ago it was found that the common soil bacterium Agrobacterium tumefaciens is capable of extraordinary feats of interkingdom genetic transfer. Since this discovery, A. tumefaciens has served as a model system for the study of type IV bacterial secretory systems, horizontal gene transfer and bacterial-plant signal exchange. It has also been modified for controlled genetic transfo...
